Most people assume a serum works simply because it is a good one, applied generously and often enough. That assumption holds up for plenty of skincare, but it breaks down completely for the two ingredients at the centre of a proper skin booster. Polynucleotides and cosmetic peptides are both built at a size skin was never designed to let through. There is a specific, well documented rule that explains exactly why.
The rule your skin already enforces
In 2000, dermatologists Jan Bos and Menno Meinardi reviewed the evidence on which chemical compounds can cross intact skin and which cannot. Their review set out what has since become known as the 500 Dalton rule. A Dalton is simply a unit of molecular weight. Their review found that molecules above roughly 500 Daltons do not passively cross the stratum corneum, the tough outer layer of dead cells that gives skin its barrier function, in any meaningful amount (Bos and Meinardi 2000). Every licensed topical and transdermal drug on the market today sits under that threshold. It is not a marketing figure. It is the working limit of what skin lets in on its own.
In short: the bigger the molecule, the less of it gets past the surface without help.
Where PDRN actually sits on that scale
PDRN, the polynucleotide fragment behind our Salmon DNA Gold booster, is a broken down piece of DNA. Even at its smallest useable fragment length, it typically sits somewhere between fifty thousand and one and a half million Daltons, depending on the source and how finely it has been processed. That is not close to the 500 Dalton threshold. It is somewhere between one hundred and three thousand times over it.
The peptide in our own booster, tested
Peptides are usually assumed to be small because the word sounds delicate, but most sit well above 500 Daltons too. Our Stayve EGF Peptide Gold booster lists Acetyl Hexapeptide-8 among its actives, better known by its ingredient trade name Argireline. In 2018, a study in Scientific Reports measured exactly what happens when that specific peptide, at 889 Daltons, is applied to skin with nothing to help it through. Only 0.22 per cent of it permeated the skin at all and just 0.01 per cent reached the epidermis (Lim et al 2018). That is not a small gap. It is the difference between a booster doing the job it is formulated for and a booster sitting on top of skin doing very little.
Why hyaluronic acid is not part of this problem
It is worth being clear that not every skincare ingredient runs into this wall. Hyaluronic acid is usually formulated across a deliberate range of molecular weights specifically so some of it can sit on the surface for hydration while smaller fractions work slightly deeper. That is a different job to what PDRN and peptide boosters are built for, which is why a hyaluronic acid serum earns its place as a standalone topical in a way a PDRN or peptide booster does not.
This is what the microchannel is actually for
Microneedling does not work by helping a formula sit better on the skin. It works by creating temporary microchannels through the stratum corneum, the exact layer the 500 Dalton rule describes. For roughly the first thirty minutes to an hour after a session those channels stay open, giving a molecule the size of PDRN or Acetyl Hexapeptide-8 a route past the barrier that no amount of massaging a serum into intact skin will create. What this means for your routine
If a PDRN or peptide booster has been sitting in your bathroom cabinet as a normal serum, it is not doing anything close to what it was formulated to do. Can I use a skin booster without microneedling?
You can apply a PDRN or peptide booster to intact skin, but at that size neither ingredient is designed to cross the stratum corneum on its own. Without the microchannels microneedling creates, a booster behaves much like an ordinary hydrating serum rather than the deeper regenerative treatment it was formulated for.
What is the 500 Dalton rule?
It is a widely cited dermatology finding that molecules over roughly 500 Daltons in weight do not passively cross intact skin in any meaningful amount. Every licensed topical and transdermal drug currently on the market falls under that threshold, which is why it is used as a practical cutoff for what skin lets through unassisted.

Does this mean my normal peptide serum is a waste of money?
Not necessarily. Peptide serums applied on intact skin can still support hydration and the skin barrier, but they do not reach the dermis in meaningful amounts without help. If firmness and collagen support is the goal, pairing a peptide booster with microneedling is what gets that specific ingredient to where it needs to be.
Do I need a Dr. Pen, or does a derma roller work too?
Either works for this. A derma roller creates the same style of microchannel as a Dr. Pen, just with a rolling motion rather than an oscillating one. It is also a lower cost way to get started. What matters for the size argument in this guide is the channel itself, not which tool creates it, so a PDRN or peptide booster applied during a derma roller session reaches the skin the same way it does with a Dr. Pen.
